چکیده

X-ray crystallography is a method used in determining the arrangement of atoms within a crystal. This method of examining a molecular structure can be applied to biological compounds, such as proteins and enzymes. By allowing researchers to know the structure of an enzyme, the function of such a molecule can be fully understood. Often, chemical properties may be known of an enzyme, but how such an enzyme truly functions is dependent upon its structure and how it interacts with its substrate and environment. Specifically, the chicken-egg white lysozyme under investigation was crystallized in order to undergo x-ray diffraction, as well as analyzed with a circular dichroism spectrometer. The growth of lysozyme crystals using varying precipitant solutions provided a study on the effects of growth and crystal lattice formation as a result of differing solutions. Concentrations were a factor changed but so were ions, which provided us with a study of the Hofmeister Series, which “ranks the relative influence of ions on the physical behavior of a wide variety of aqueous processes” (Zhang). The data collected validated the Hofmeister Series and provided the conclusion that the precipitant present in the buffer solution does affect the size and shape of the crystal grown.

Enhancing protein crystallization through precipitant synergy.

Suitable conditions for protein crystallization are commonly identified by screening combinations of independent factors that affect crystal formation. Because precipitating agents are prime determinants of crystallization, we investigated whether a systematic exploration of combinations of mechanistically distinct precipitants would enhance crystallization. A crystallization screen containing ...

Forward-Light-Scattering Characterization of Pre-Crystalline Aggregates in Crystallizing Lysozyme Solutions

We present a method to characterize lysozyme pre-crystalline aggregates using a forward-lightscattering technique, which is highly sensitive to protein aggregates. The static light scattering properties at small angles of crystallizing lysozyme aggregates are discussed, and related to the crystallization conditions based on the concentration of added precipitant NaCl. Lysozyme solutions that st...

Protein crystal growth. Growth kinetics for tetragonal lysozyme crystals.

A method for immobilizing protein crystals has been devised for determining face growth rates, and used to investigate the growth kinetics of hen egg white lysozyme crystals. Growth rates were determined at 22 degrees C in 0.1 M sodium acetate, 5% NaCl, pH 4.0, on the visually identified (110) face of tetragonal lysozyme crystals. Protein concentrations ranged from 13 to 57 mg/ml (saturation co...

Growth of wurtzite ZnO nanorods using different capping agents: Characterization, morphology, and investigation the catalytic activity in some oxindoles and indolyl organics

ZnO nanorods have been prepared through chemical deposition of Zn(OAc)2.2H2O by employing different capping agents, (PEG, MW=2000 and PEG, MW=5000). The fabricated catalyst was characterized by scanning electron microscope (SEM) images and XRD pattern. The results show the one dimensional growth of ZnO nano-rods. The results show that capping agents can control the shape and growth of nano-size...
